SERD stands for Selective estrogen receptor degrader, a drug class abbreviation.

Hormonal breast cancer drugs that bind the estrogen receptor and cause its destruction, used for hormone receptor-positive advanced breast cancer. Fulvestrant is given by monthly intramuscular injection; elacestrant was the first oral SERD.

Example: ESR1 mutation detected on liquid biopsy: switch to oral SERD elacestrant.

Usage notes

Fulvestrant was approved in 2002 and is given as two 5 mL injections, one in each buttock. Elacestrant (Orserdu), approved in 2023, is indicated for tumors with ESR1 mutations, which commonly develop after aromatase inhibitor therapy and make cancers less responsive to estrogen deprivation. Several other oral SERDs are in development.
