sgRNA stands for Single guide RNA, a genetics abbreviation.

Engineered RNA that directs the Cas9 enzyme to a chosen DNA target in CRISPR gene editing. Its first 20 nucleotides match the target site, and the rest forms a scaffold that binds Cas9; designing an sgRNA is often the first step of an editing experiment.

Example: Three sgRNAs targeting exon 2 were tested, and the most efficient one was used for knockout.

Usage notes

Also called guide RNA (gRNA). Natural CRISPR systems use two separate RNAs, crRNA and tracrRNA; Martin Jinek, Jennifer Doudna, Emmanuelle Charpentier and colleagues fused them into a single chimeric guide in 2012. Online design tools score sgRNAs for predicted efficiency and off-target risk.
