JAK2 stands for Janus kinase 2, a hematology and oncology abbreviation.
Signaling enzyme that relays growth signals from cytokine receptors inside blood-forming cells. The JAK2 V617F mutation, discovered in 2005, is present in about 95 percent of polycythemia vera and about half of essential thrombocythemia and primary myelofibrosis.
Example: JAK2 V617F detected at 42% allele burden, supporting a diagnosis of PV.
Usage notes
Testing for JAK2 is the first step when a myeloproliferative neoplasm is suspected, followed by CALR and MPL if negative. JAK inhibitors such as ruxolitinib treat myelofibrosis, polycythemia vera, graft-versus-host disease and some inflammatory conditions. JAK2 mutations can also appear in apparently healthy older people as clonal hematopoiesis.
