LEN stands for Lenacapavir, a drug abbreviation.
First-in-class HIV capsid inhibitor given as an injection under the skin every six months. It is approved for multidrug-resistant HIV (Sunlenca) and as twice-yearly pre-exposure prophylaxis (Yeztugo).
Example: PrEP: LEN 927 mg subcut every 6 months, with oral loading tablets on days 1 and 2.
Usage notes
In the PURPOSE 1 trial, reported in 2024, no HIV infections occurred among young women in South Africa and Uganda receiving lenacapavir. The FDA approved it for PrEP in 2025. Its long interval could greatly improve adherence compared with daily pills, though cost and global access remain debated.
