1. Anti-double-stranded DNA antibody

Medical · Laboratory Test Abbreviations

Autoantibody highly specific for systemic lupus erythematosus, positive in roughly 60 to 70 percent of people with the disease. Rising levels, especially with falling complement C3 and C4, often signal a flare and are associated with lupus nephritis.

Example: dsDNA up to 340 IU/mL with low C3; urine protein ordered to screen for nephritis.

Methods include ELISA, the Crithidia luciliae immunofluorescence test and the older, very specific Farr radioimmunoassay, and results are not interchangeable between methods. Drug-induced lupus from hydralazine or procainamide usually produces anti-histone rather than anti-dsDNA antibodies, a useful distinction. Often written anti-dsDNA.

2. Double-stranded DNA

Science & Engineering · Genetics Abbreviations

DNA in its usual form of two complementary strands paired into a double helix. Lab protocols specify dsDNA when measuring DNA concentration, describing virus genomes or testing for autoimmune antibodies.

Example: Quantify the library with a dsDNA-specific fluorescent assay rather than UV absorbance.

Many viruses, such as herpesviruses and adenoviruses, have dsDNA genomes. Antibodies against dsDNA are a characteristic laboratory finding in systemic lupus erythematosus. At an absorbance of 1.0 at 260 nm, a solution contains about 50 nanograms per microliter of dsDNA.
